Repare Therapeutics는 Versant Ventures가 Inception Therapeutics의 Discovery Engine을 통해서 18개월간 stealth mode로 “CRISPR-enabled synthetic lethality drug discovery platform”을 통해서 oncology targets와 새로운 프로그램을 발굴했고 이것을 전임상으로 이끌기 위해 $68 Million의 Series A를 했습니다.

Co-Founders는 Daniel Durocher, Ph.D., Agnel Sfeir, Ph.D., 그리고 Frank Sicheri, Ph.D.입니다.

• Dr. Sfeir from the Department of Cell Biology and the Skirball Institute of Biomolecular Medicine at NYU Langone Medical Center

• Drs. Durocher and Sicheri from Toronto’s Lunenfeld-Tanenbaum Research Institute

Repare의 첫번째 프로그램은 “DNA-directed DNA polymerase theta (PolQ)”라고 하는 암세포에서 DNA double-strand break를 repair하는 중요한 단백질입니다. PolQ IP는 NYU Dr. Sfeir의 연구결과를 인수한 것입니다.

그리고 2년후에 PolQ program은 일본의 ONO Pharmaceuticals와 전략적 제휴를 하고 그 이후 시리즈 B를 했고요.

Series B를 한 이후 발표한 내용을 보면 ATR inhibitor RP-3500 (Camonsertib)의 임상시험을 후년에 하는 것이 계획이었습니다. 그리고 2022년에 Roche와 upfront $125 Million 포함 총 금액 $1.2 Billion 규모의 계약을 통해 전략적 제휴를 맺었습니다.

그리고 첫번째 마일스톤으로 $40 Million을 Roche로 부터 받았습니다.

Camonsertib (RP-3500)의 임상 1상 결과는 Nature Medicine 에 발표를 했는데 결과는 좋습니다.

그런데 사실상 Registrational study라고 할 수 있는 phase 2를 시작한지 열흘만에 Roche가 갑자기 이 약물의 권리를 포기한다는 발표를 했습니다.

이런 갑작스런 Roche의 발표와 그 배경에 대해 아직 아무도 해설을 내지 못하고 있는 실정입니다.

Camonsertib (RP-3500)과 PKMYT1 inhibitor인 Repare의 Lunresertib (RP-6306)의 병용 임상1상 데이터도 발표한 자료에 의하면 매우 좋은 상태여서 Camonsertib의 문제는 아닌 것은 확실한 것 같습니다.

ATR inhibitor의 임상 경쟁상황이 매우 높은 상황인데 AstraZeneca의 Ceralasertib (AZD6738)가 현재 임상 3상을 Duralumab과 병용으로 진행하고 있습니다.

이외에도 Merck/Vertex (Berzosertib), Bayer (Elimusertib) 등이 임상 진행 중이며 가장 최근에는 Merck KgaA의 Tuvusertib (M1774)의 초기 임상결과 안정성이 좋은 신물질임을 발표했습니다.

Pol theta program의 첫번째 물질로 ONO Pharmaceuticals와 개발 중이던 RP-2119는 2023년 7월 31일 계약이 종료되었고 후속물질인 RP-3467이 IND-enabling study를 진행 중입니다.

올해 1월에 발표한 Repare Therapeutics의 Presentation을 보면 현재 자금력은 $223 Million으로 2026년까지 무리없이 운용할 수 있는 상황이고 파이프라인도 비교적 잘 진행되고 있습니다.
