Antibody-Drug Conjugate (ADC)는 Antibody의 Cell Specificity와 Small Molecule Toxin의 Cytotoxicity를 Bioconjugation을 통해 연결해서 암세포 등을 죽이는 Targeted Oncology Therapeutics인데요 ADC회사인 레고켐에 대해 블로그를 쓴 적이 있습니다.

Yale University의 Craig Crews 교수 연구실의 연구결과로 시작된 Targeted Protein Degrader (TPD)와 Molecular Glue 등을 이용해서 특정 단백질을 분해시키는 Small Molecule을 항암제로 개발하는 회사들도 많이 있습니다. 예를 들면 Arvinas 가 있죠.
ADC와 TPD를 결합한 버전인 약물이 Degrader-Antibody Conjugate (DAC)이라는 Platform Technology를 배경으로 새로 탄생한 회사가 있습니다.

1년전에 Versant Ventures의 스위스 바젤의 인큐베이터인 Ridgeline Discovery Egine을 통해서 Firefly Bio를 설립했고 이번에 Series A를 한 것입니다. Versant Ventures의 California에 있는 Discovery Engine인 Inception Therapeutics처럼 스위스에는 Ridgeline Discovery Engine이 있습니다.

Versant Ventures와 MPM BioImpact가 Series A $94 Million를 주도했는데요 Nobel Prize Winner인 Carolyn Bertozzi교수와 Genentech/Merck veteran인 Scott Hirsch를 CEO로 영입해서 Firefly Bio를 설립했습니다. CSO로는 Genentech과 Merck에서 ADC 연구한 John Flygare 박사와 Bernhard Geierstanger박사는 Novartis와 Merck에서 ADC 개발을 한 베테랑으로서 CTO로 역할을 합니다. Firefly는 이 자금을 통해서 DAC Platform을 발전시킬 예정인데요 현재 15명의 임직원을 연말까지 40명까지 증원한다는 계획입니다.
“Over the last 20 years ADCs, have been dominated by toxins, and what we’re trying to do at Firefly is introduce a new payload class with the same level of potency as toxins, but that can do a better job discriminating healthy from diseased tissue,” the new CEO said in an interview.
At Firefly, we’ve assembled a unique talent base and platform to significantly advance this promising field,” said Jerel Davis, Ph.D., Managing Director at Versant and a Firefly board member. “The company has made rapid progress and is poised to become the leader in discovering clinically meaningful DACs in oncology and immunology.”
However, Davis admitted that some recent deals have stepped into the biotech’s lane. 최근에 DAC에 대한 Big Pharma의 관심으로 몇가지 Deal이 있었습니다.

Firefly Bio는 Solid tumor target에 대해 DAC를 적용할 계획을 가지고 있고 후보약물도 있다고 밝혔습니다. 이번 증자에는 Eli Lilly가 함께 참여했는데요. 아직도 Stealth-mode라서 더 자세한 것은 밝히지 못하지만 곧 알 수 있으리라 기대합니다.
Carolyn Bertozzi 교수는 LYTAC이라는 Protein Degrader를 발견하고 2021년에 Nature Chemical Biology에 논문을 냈고요.
2023년에 Science에 LYTAC에 대한 새로운 논문을 발표한 바 있습니다.
LYTAC을 통한 DAC Platform을 개발하고 있는 것으로 보입니다. 아래는 Carolyn Bertozzi교수께서 작년에 발표하신 LYTAC에 대한 세미나 영상입니다. LYTAC을 이용한 다양한 DAC program 개발은 새로운 영역을 여는 것임에 틀림없습니다. 좋은 신약이 나올 수 있기를 바랍니다.
