안녕하세요 보스턴 임박사입니다.
바이오텍에 대한 블로그를 쓰는 중에 제가 사는 곳이 보스턴이다 보니 너무 지역적인 한계 속에서 미국 위주의 바이오텍에 대해 글을 쓰고 있지 않는지 반성을 하게 되네요. 오늘은 독일의 바이오텍 회사에 대해서 얘기를 좀 해 보려고 합니다. 회사는 Ethris GmbH라는 회사인데요 이 회사는 독일 남부 뮌헨 근처에 있는 회사입니다. 2009년에 회사가 설립된 이후에 계속해서 연구 위주의 노력을 해 온 것으로 보입니다.
이 회사는 Ludwig Maximilian’s University Munich 소아과 교수인 Carsten Rudolph박사의 2011년 Nature Biotechnology에 보고한 Modified mRNA 기술을 토대로 하고 있습니다.
2017년 Molecular Therapy에 ACE2 mRNA의 간 및 폐 전달에 대한 쥐실험 결과를 보고했습니다.
Ethris와 공동으로 뮌헨 대학교팀에서 Plos One 2015에 발표한 논문이 있는데요 이것을 보면 Lipidoid 가 PEI인 것을 알 수 있습니다.
Ethris의 2015년에 공개된 미국 특허에서도 PEI 기반 전달체를 사용하고 있습니다.
그러다가 2017년에 Astrazeneca와 5년간 계약을 맺으면서 $29.5 Million upfront payment를 포함한 연구자금을 받게 되었습니다.
“Through the agreement, Ethris will work exclusively with AstraZeneca and its MedImmune and Innovative Medicines (IMED) units to develop multiple targets for evaluation against asthma, chronic obstructive pulmonary disease, and idiopathic pulmonary fibrosis. Ethris will receive a €25 ($29.5 million) million up-front payment plus research funding. The firm is also eligible to earn potential R&D milestones and sales-related royalties. AstraZeneca and MedImmune retain the option to exclusively license each target within the collaboration upon completion of the agreed research plan. “
2022년 초에 독일 VC인 Laureus Capital을 통해서 $26 Million 규모의 시리즈 B를 성공적으로 마쳤습니다. 이 자금을 가지고 ETH47 (a broad anti-viral immune modulator)과 ETH42 (a protein-replacement therapy for primary ciliary dyskinesia (PCD))의 전임상과 임상을 할 자금을 확보했습니다. Ethris는 Lipidoid Nanoparticle 을 전달체로 사용합니다. 이것은 Lipid Nanoparticle (LNP)와는 다른 형태의 나노입자로서 아직 임상에서 사용되지 않은 기술입니다.
Ethris의 첫번째 DC (Drug candidate)인 ETH47은 Type III interferon mRNA drug인데요 호흡기를 통한 전달기술로 mRNA를 전달하는 약물입니다. Covid-19 pandemic이 거의 마쳐진 2023년 11월말에 영국에서 처음으로 임상을 시작했습니다.

“ETH47 is a type III interferon (IFN)-encoding mRNA. The drug induces an innate immune defence response in the mucosa at virus entry sites. It can also inhibit viral replication, with Ethris stating the drug has the ability to address a broad range of respiratory virus infections.”
Ethris는 Lonza와 Spray-dried mRNA vaccine formulation을 개발하는 협약을 했습니다. Lonza가 인수한 Bend는 formulation 분야에서 특화된 회사입니다. 이 회사를 통해서 좋은 spray dry formulation을 개발하고자 하는 결정은 좋은 결정이라고 생각합니다.
” The data indicates that ETH47, administered nasally, was generally safe and well-tolerated, with no severe adverse events or drug-related adverse events of clinical relevance across all dose levels. Initial results also show target engagement and suggest no systemic biodistribution of ETH47. A full data readout is planned for the third quarter of 2024.”

현재 Ethris의 pipeline은 아래와 같습니다. 현재 ETH47이 천식 (Asthma) 환자를 대상으로 임상2상을 진행 중이고 COPD 환자를 대상으로 임상 1상을 진행 중입니다.
Ethris의 CEO인 Carsten Rudolph와 CMO인 Thomas Langenickel이 인터뷰한 Video가 있는데요 링크를 아래에 걸었고요. Ethris의 mRNA-lipidoid nanoparticle의 Pulmonary delivery platform이 어떤 결말을 맞을지 궁금합니다.
제가 보기에는 Lipid Nanoparticle (LNP)로 전달체를 수정해야 하지 않을까 하는 생각이 듭니다. 하지만 항상 저의 생각보다 더 좋은 임상 결과를 볼 수 있기를 항상 기다립니다.
