안녕하세요 보스턴 임박사입니다.
매주 수요일은 일하는 주의 중심에 있어서 그런지 수요일은 뭔가 한주의 축이 넘어가는 정점에 있는 지렛대와 같은 느낌이 듭니다. 얼마전에 수요일에 출근을 하는데 정말이지 너무 길이 막히고 어렵게 회사에 도착을 했는데 주차장에 자리가 거의 없어서 간신히 차를 주차했습니다. 그리고 나서는 수요일은 웬만하면 재택근무를 하게 되었습니다. 오늘 미팅을 하다보니 저와 비슷한 생각을 가진 사람들이 꽤 되는 것 같더군요.
한동안 항체 관련한 글을 올리지 못했는데 제가 이 분야에 대해 여전히 배움이 부족하다는 생각이 많아서 그런건지 눈에 잘 들어오지 않는 면이 있습니다. 그래도 혹시 자꾸 배우다 보면 서당의 개가 풍월을 읊을 수 있듯이 뭔가 잘 모르더라도 안목이라도 생길 수 있지 않을까 하는 마음으로 항체치료제에 대한 회사를 공부해 보려고 합니다.
오늘 얘기할 회사는 Deren Bio라는 회사인데요 2019년에 Dr. Nenad Tomasevic라는 세르비아 출신 과학자가 창업한 회사입니다. Dr. Nenad Tomasevic는 세르바이에서 박사학위를 하고 미국 NIDDK에서 포스닥을 한 이후에 줄곧 바이오텍에서 일을 한 바이오텍 스타트업 베테랑입니다.

Dren previously closed a $6 million Seed Financing in 2019 led by 8VC with participation by Mission BioCapital and prominent industry veterans…Dren Bio, Inc., a company developing powerful protein-engineering technologies for depletion of cells, protein aggregates and other disease-causing agents, announced the closing of a $60 million Series A investment round. The round was led by SR One and Taiho Ventures, LLC , with participation from existing investors 8VC and Mission BioCapital and new investors including BVF Partners L.P., HBM Healthcare Investments, and Alexandria Venture Investments.
2019년에 8VC 등으로 부터 Seed rounding $6 Million을 하고 DR-01과 DR-02를 개발했습니다.
그리고 2년 후에 시리즈 B와 함께 Pfizer와 딜을 합니다.

Though Pfizer is a new investor, the Big Pharma partnered with Dren at the beginning of this year to develop therapeutic bispecific antibodies for certain oncology targets. Dren received $25 million upfront as part of the deal, which is valued at more than $1 billion in potential cash payments plus potential product sales royalties.
$65 M 시리즈 B와 함께 Pfizer로 부터 $25 M upfront를 포함한 총 규모 $1 Billion의 연구계약을 체결했고 DR-01의 임상1상을 준비하고 있었습니다.
창업한지 5년 정도가 된 후에 비로소 어떤 플랫폼인지가 알려집니다.
Dren Bio’s Targeted Myeloid Engager and Phagocytosis Platform
이 플랫폼에 노바티스에서 $150 Million의 upfront, $25 Million 지분투자를 포함한 총 규모 $2.85 Billion의 연구계약을 체결했습니다. Dren Bio는 Drug Candidate를 개발하고 그 이후 개발부터 생산, 임상, 상용화까지는 노바티스가 맡는 조건의 계약이었습니다.
Dren Bio’s lead asset is DR-01, which targets autoreactive CD8 T cells and is currently in phase 2 trials for cytotoxic lymphomas. The biotech’s platform is designed to activate myeloid cells by engaging a phagocytotic receptor that is only expressed on those cells.
DR-01은 CD8 T cells를 표적으로 하고 cytotoxic lymphoma에 대해 임상 2상을 진행하고 있었습니다. 이 회사의 플랫폼은 phagocytotic receptor를 통해서 myeloid cells를 활성화하는 것으로 Myeloid Cell Engager (MCE)라고도 합니다.
그리고 올해 3월에 Sanofi가 $600 Million upfront를 포함한 $1 Billion 딜로 CD20-directed antibody (DR-0201)이라는 bispecific myeloid cell engager를 확보했습니다. 현재 이 약물은 B-cell non-Hodgkin lymphoma의 임상 1상을 진행 중인데 임상 결과는 공개되지 않았지만 “robust” B-cell depletion을 확인했다고 사노피가 공시했습니다.

The CD20-directed antibody, dubbed DR-0201, is a bispecific myeloid cell engager Dren has been evaluating in a phase 1 trial for B-cell non-Hodgkin lymphoma. While the biotech has yet to announce any clinical results for the antibody, Sanofi implied in a March 20 release that it’s pleased with the “robust” B-cell depletion it has seen so far.

Targeted Myeloid Engager and Phagocytosis Platform
Dren Bio’s Targeted Myeloid Engager and Phagocytosis Platform is a bispecific antibody-based technology that induces potent depletion of pathogenic cells, protein aggregates, and other disease-causing agents by engaging a novel phagocytic receptor that is selectively expressed on myeloid cells. Bispecific antibodies generated from the platform are specifically engineered to enable controlled myeloid cell activation only in the presence of the target antigen, which may result in greater therapeutic indexes and offer superior safety profiles compared to other therapeutic modalities such as T-cell engagers and Antibody Drug Conjugates (ADCs). The platform is being deployed across various therapeutic areas and has generated multiple bispecific antibody programs to address unmet need in oncology, immunology, and neurology.
현재 아주 다양한 파이프라인이 있는데 주로 파트너쉽을 통해서 개발부터 상용화까지는 빅파마가 하고 Dren Bio는 플랫폼 개발에 집중하는 것으로 보입니다.

5-6년만에 정말 대단한 플랫폼의 성장 속도와 파트너쉽 속도입니다. 올해 상을 받았군요. 지금보다 향후 미래가 더욱 주목이 됩니다. 이 정도의 파트너쉽 모델이라면 IPO하지 않아도 될 것 같습니다. 기대가 많이 되는군요.
